Fiber type distribution, succinate dehydrogenese and peroxisome proliferator- activated receptor fÁ-coactivator-1f¿ gene expression level of fibers in the soleus muscle of Zucker diabetic fatty rats

   Abstract
We have reported that fiber type distribution change is detected in two type diabetic model rats (OLETF and GK rats). In this study, we determined whether the soleus muscle fiber type distribution change was caused by diabetes, using obese and diabetic (Zucker diabetic fatty, ZDF), obese but non-diabetic (Zucker fatty, ZF), and non-diabetic and non-obese (Zucker lean, ZL) rats. Moreover, we investigated whether the expression levels of metabolic key molecules, the transcriptional factor of metabolic genes, such as peroxisome proliferator-activated receptor fÁ coactivator-1f¿ (PGC-1f¿) and the oxidative enzyme in mitochondria, such as succinate dehydrogenase (SDH), were changed in type I and II muscle fibers in each rat, using new technique, laser capture microdissection (LCM). Both plasma glucose and HbA1c levels were significantly higher in ZDF than ZL and ZF rats. A lower percentage of type IIA fiber was observed in the muscle of ZDF rats than in those of ZL and ZF rats. The mRNA expression levels of SDH in type II fibers and of PGC-1f¿ in type I fibers were significantly lower in ZDF than in ZL and ZF rats using LCM and real-time PCR analysis. We first clarified that a lower percentage of type IIA fibers was observed in diabetic and obese ZDF rats. We also discovered that the expression level of oxidative metabolic related genes, PGC-1f¿ and SDH, decreased in type I and type II fibers, respectively, of ZDF rats.
